Bradycardia induced by Mesobuthus tamulus scorpion venom involves muscarinic receptor-G-protein-coupled cell signaling pathways.

چکیده
Indian red scorpion (Mesobuthus tamulus; MBT) envenomation produces various cardio-respiratory abnormalities including cardiac dysrhythmias. The underlying cell signaling pathways for the cardiac dysrhythmias produced by MBT venom are not known. The present study was therefore conducted to delineate the second messenger signaling pathways involved in MBT venom-induced atrial rhythm changes. The effects of venom and various antagonists were examined on spontaneously beating rat right atrial preparations in vitro. The MBT-venom produced an increase (35%), a decrease (45%) and again an increase (50%) in rate at 0.03, 0.3 and 3.0 microg/ml of venom, respectively. On the other hand, force of contraction exhibited a concentration-dependent rise (up to 40%) at all concentrations of venom. Pretreatment with atropine (0.3 microM) blocked the decrease in atrial rate at 0.3 microg/ml concentration of venom while no such blockade was seen in force of contraction. Submaximal concentration of ACh (0.1 nM) decreased the atrial rate by 25%. In the presence of MBT venom (0.3 microg/ml), ACh-induced fall in atrial rate was enhanced. The venom-induced fall in atrial rate and augmentation of ACh response were blocked by pertussis toxin (PTx; a Gi-inhibitor) or methylene blue (a G-cyclase inhibitor). The results indicate that the decrease in atrial rate produced by venom is mediated muscarinic by receptors via Gi-guanylyl cyclase mediated cell signaling pathways.

Studies on Antivenom Activity of Aristolochia indica Plant Extract Against Red Scorpion Venom by in Vivo and in Vitro Methods
Objective: Mesobuthus tumulus is the most lethal among all poisonous species of scorpion. Aristolochia indica is one of the plants which have long been used in traditional herbal medicine for the treatment of poisoning by animal bites. Hence the study was planned to evaluate the ethanolic extract of Aristolochia indica for the treatment of Mesobuthus tamulus envenoming. Materials and Methods: C...
متن کاملIsolation of 5-Hydroxytryptamine from the Venom of the Indian Red Scorpion (Mesobuthus tamulus)
The present study was undertaken to isolate 5-hydroxytryptamine (5-HT) from crude Indian red scorpion (Mesobuthus tamulus; MBT) venom as it has not been reported yet. Therefore, with the help of bioassay (using gastric fundus contractions) and fluorimetric assay methods, the 5-HT content of crude MBT venom (CV) was determined. The concentration of 5-HT in CV by either of the methods was differe...
متن کامل2-Deoxy-D-glucose reverses the Indian red scorpion venom-induced cardiopulmonary abnormalities in anesthetized rats.
Role of 2-Deoxy-D-glucose (2-DG) in reversing the Indian red scorpion (Mesobuthus tamulus concanesis Pocock, MBT) venom-induced toxicity was examined. Femoral arterial pressure, ECG and respiratory movements were recorded in urethane anesthetized rats. Plasma glucose and serum insulin levels were also estimated. Intravenous injection of 5 mg/kg MBT venom produced immediate decrease in mean arte...
متن کاملIncreased osmotic fragility of red cells in dogs with acute myocarditis produced by scorpion (Buthus tamulus) venom.
Stings from scorpions (Buthus tamulus) produce acute myocarditis and can result in death in children and adults. Acute myocarditis was induced in anaesthetised dogs by intravenous injection of 4 mg/kg venom (Buthus tamulus). Myocarditis was confirmed by ECG. Blood was collected before and 30 minutes after venom treatment and processed for osmotic fragility. An increase in osmotic fragility of r...
متن کاملPathophysiological approach to the management of scorpion envenomation.
Indian red scorpion (Mesobuthus tamulus; MBT) produces lethal stings and is a matter of concern in certain parts of India. MBT envenomation produces multi-systemic involvement, thus presents difficulty in the management. Symptomatic treatment has been practiced earlier that failed to relieve the toxic effects of the venom. Therefore, present manuscript deals with pathophysiologically based appr...
